PARACHINAR, Pakistan (AP) ? A suicide bomber on a motorcycle detonated his explosives near a vehicle carrying a candidate from a hard-line Islamist party in northwest Pakistan on Tuesday, killing 12 people in the second attack on the party in as many days, police said.
No one has claimed responsibility for the bombing, but the Taliban took credit for an attack the day before on the same party, claiming it was targeting a candidate who had supported military operations against the militants in the northwest.
The blast Tuesday also wounded 35 people, but the candidate from the Jamiat Ulema-e-Islam party, Mufti Syed Janan, escaped unharmed, said police officer Haleem Khan. The attack occurred as Janan's convoy passed through a market in the town of Doaba in northwest Khyber Pakhtunkhwa province, said Khan.
The Pakistani Taliban set off a bomb at a political rally held by Jamiat Ulema-e-Islam in the northwest Kurram tribal region on Monday, killing 25 people and wounding 70, said government official Javed Khan. The targeted candidate was not harmed.
The Taliban have carried out multiple attacks in the run-up to national elections scheduled for May 11. But most of the attacks have targeted secular parties that have opposed the militants and backed the army's attempt to clear them from their sanctuaries in the northwest.
Prior to the last two days' bombings, there was concern that the attacks could benefit parties that take a softer line toward the militants, like Jamiat Ulema -e-Islam, because their candidates are able to campaign more freely ahead of the vote.
But the Taliban have also condemned democracy as a whole, meaning that any political party taking part in the elections could be considered fair game by the militant group. Militants have called on people in many areas to stay away from the polls on election day.
The Jamiat Ulema-e-Islam party is considered supportive of the Afghan Taliban's fight against the United States and its allies in neighboring Afghanistan. It's also sympathetic to the Pakistani Taliban, which have been fighting Pakistani troops and would like to establish a hardline Islamic government in Pakistan. The group's leaders have generally opposed the Pakistani military's operations against the militants and instead called for negotiating with them.
But that hasn't made the group immune.
In 2011, a suicide bomber struck a convoy in which the party's head, Maulana Fazlur Rehman, was traveling through northwestern Pakistan, killing 12 people.

NEW YORK (Reuters) - Antero Resources, an oil and natural gas company controlled by Warburg Pincus LLC, is preparing for an initial public offering that could value it at as much as $10 billion, three people familiar with the matter said on Monday.
The Denver, Colorado-based company has hired Barclays Plc
Barclays, Citigroup and Warburg declined to comment. JPMorgan could not be reached for comment.
Antero reported adjusted earnings before interest, tax, depreciation and amortization of $434.3 million in 2012, up from $340.8 million in 2011.
The company has around 311,000 acres in the Marcellus Shale formation in West Virginia and Pennsylvania as well as 92,000 acres in Ohio's Utica Shale.
Antero believes that most of their acreage in the Marcellus holds natural gas liquids, which tend to be worth more than "dry" gas.
The company said its Utica acreage sits in a region that it believes holds natural gas liquids. The Utica shale is an emerging region that has attracted the interest of big oil companies like Exxon Mobil Corp
Antero's said earlier this year that its proved, probable and possible reserves were 26.1 trillion cubic feet equivalent of gas and oil.
According to Thomson Reuters data, there have been only three U.S.-listed IPOs in the energy and power space year-to-date that raised proceeds of $973 million. That's down from eight companies which raised $2.2 billion during the same time last year.
Warburg and other investors including energy-dedicated private equity firm Yorktown Energy Partners and Lehman Brothers Merchant Banking Group initially invested $260 million in Antero in 2003. Four years later, the consortium led a $1 billion equity investment in the company.
Private equity firms are increasingly looking to public markets in 2013 to profitably exit their investments as the frothy stock market has buoyed the value of companies public and private alike.

Worried? Don't be. Even at their worst, they're more bothersome than troublesome.
They are noisy, though. To attract a mate, males push air though vibrating organs called tymbals to create a whiney-hissy sound. A tree full of horny males can reach 90 decibels when measured directly beneath, which is comparable to a lawn mower. However, that's unlikely to cause hearing damage in humans; you'd need continuous exposure for it to pose a risk. In case you were curious, OSHA workplace standards allow for up to eight hours of continuous noise at that level.
The Humane Society warns against allowing pets, especially dogs, to gorge on a yard full of cicadas. Aside from being a choking hazard, the bugs have hard shells made of chitin, and in large amounts it clogs up digestive systems and causes constipation and vomiting. Cicadas treated with pesticides can also be toxic to pets.
Most of the damage is caused by egg-laying females. Using their ovipositors, they make tiny slits in young branches before depositing around 25 eggs and moving on. They may repeat the process as many as 20 times in a single twig. Although a single cicada won't do much harm, heavy infestations can cause young branches to bend over or "flag." Damage to mature trees is rarely serious, but younger trees and saplings can suffer, so protect them with 1/4-inch mosquito netting. Generally, avoid planting new trees during the spring of a known brood emergence.
Female cicadas will target trees that provide the best food for the nymphs, such as fruit trees, birches, oaks, maples, and hickory, and they prefer to lay their eggs in branches 1/2 inch in diameter or smaller. Generally, they avoid most other plants such as ornamental shrubs, flowers, herbs, and vegetables, as well as pine and fir trees.
While underground, nymphs suck xylem fluids from the tree's roots. Prolonged feeding can hamper plant growth and fruit production.
Cicadas will stay clear of the house. In other words, they won't try to burrow into its eaves or lay eggs in the siding.
Don't gorge on them unless you want painful stomach cramps. Also, cicadas are arthropods, so stay away if you're allergic to shrimp, lobster, or other similar seafoods. Finally, avoid them if you think they've been around garden chemicals such as pesticides and fertilizers.
Otherwise, You can mulch their shells in the garden. The chitin will help retain moisture in the soil and fend off weeds. Once the cicadas begin to decay, they make excellent fertilizer?just let nature take over and allow soil microorganisms and other bugs return the nutrients back into the soil. Also, you can use dead cicadas as fish bait.

MOSCOW (AP) ? Secretary of State John Kerry is making his case to Russian President Vladimir Putin for Russia to take a tougher stance on Syria at a time when Israel's weekend air strikes against the beleaguered Mideast nation have added an unpredictable factor to the talks.
Kerry arrived Tuesday in Moscow for talks with the most powerful ally of Syrian President Bashar Assad's regime.
Officials said Kerry hopes to change Moscow's thinking on Syria with two new angles: American threats to arm the Syrian rebels and evidence of chemical weapon attacks by the Assad regime.
Over the weekend, Israeli warplanes targeted what Israel claimed were caches of Iranian missiles bound for Hezbollah, the Lebanon-based terror group. Such weapons would allow Hezbollah to strike Tel Aviv and as far as southern Israel from inside Lebanese territory.
Israel's willingness to hit Syrian targets it sees as threats to its own existence has complicated the Obama administration's internal debate over what to do about Syria.
Israel's actions put Damascus and Moscow on notice that the U.S. and its allies may not wait for an international green light to become more actively engaged in the Syrian conflict. The administration said last week it was rethinking its opposition to arming the Syrian rebels or taking other aggressive steps to turn the tide of the two-year-old civil war toward the rebels.
At the same time, Israeli involvement in the war carries risks. Instead of prodding Russia into calling for Assad's ouster, it could bring greater Arab sympathy for Assad and prompt deeper involvement from Iran and Hezbollah, actors committed as much to preserving Assad as to fighting the Jewish state.
Although Israel hasn't officially acknowledged it carried out the airstrikes, Syrian officials on Monday were blaming Israel, calling them a "declaration of war" that would cause the Jewish state to "suffer."
Russia, alongside China, has blocked U.S.-led efforts three times at the United Nations to pressure Assad into stepping down.
U.S. officials are hoping Syria's behavior could shift Russia's stance.
"We have consistently, in our conversations with the Russians and others, pointed clearly to Assad's behavior as proof that further support for the regime is not in the interest of the Syrian people or in the interest of the countries that have in the past supported Assad," White House spokesman Jay Carney said.
U.S. officials said the administration doesn't believe the weekend activity will force President Barack Obama's hand, noting that the main U.S. concern is the use of chemical weapons by Assad, while Israel's top concern is conventional weapons falling into the hands of its enemies.
The chemical weapons argument is now under surprising attack, with former war crimes prosecutor Carla Del Ponte saying over the weekend she and fellow members of a four-member U.N. human rights panel have indications the nerve agent sarin was used by Syrian rebel forces, not by government forces.
That theory was rejected by U.S. officials. The State Department said the administration continues to believe that Syria's large chemical weapons stockpiles remain securely in the regime's control.
The Obama administration opened the door to new military options in Syria after declaring last week it strongly believed the Assad regime used chemical weapons in two attacks in March. Two days after that announcement, Defense Secretary Chuck Hagel said arming the Syrian rebels was a policy consideration.
Until now, U.S. efforts to bolster the rebels' fighting skills and gather intelligence on the groups operating inside Syria have been limited to small training camps in Jordan, according to two U.S. officials who weren't authorized to speak about secret activities and demanded anonymity.
There are several options for escalation, ranging from arming the rebels to targeted airstrikes and no-fly zones. However, arming the rebels is the most likely escalation, officials said.
While the Israeli actions have made Kerry's Russia efforts more unpredictable, some in Congress tried to be optimistic.
Maryland Rep. Dutch Ruppersberger, the top Democrat on the House Intelligence Committee, said he hopes Kerry can persuade Russia to use its influence to convince the besieged Syrian leader that he should step down.
"Hopefully the cooperation on the (Boston) Marathon bombing will open the door there," Ruppersberger said.
After visiting Moscow for the first time since he became secretary of state, Kerry will travel to Rome for talks with members of the new Italian government, as well as meetings with Israeli Justice Minister Tzipi Livni and Jordanian Foreign Minister Nasser Judeh to discuss Middle East peace prospects.

A team of zoologists have been looking for the Formosan clouded leopard, indigenous to Taiwan, for 13 years. Last week they announced it was extinct.
The news that the clouded leopards that once prowled the lowlands of Taiwan have become extinct here has prompted expressions of concern among an increasingly conservationist public.

The Formosan clouded leopard, a subspecies of large cats, probably died out here some 100 years ago, Taiwan university researchers determined after a four-year study found no photos, fur, or paw prints in a preserve considered their most likely home.
The news has led many Taiwanese to question the side-effects of the island?s economic development. In turn, animal advocates say this examination has fueled conservation movements. ?
?A lot of people have said they are disappointed and find our discovery quite regrettable,? says Kurtis Pei, a study leader from National Pingtung University of Science and Technology?s Institute of Wildlife Conservation. ?Some say they hope not just to feel regret but to do something to save other animals.?
Mr. Pei and five other researchers set up cameras and catnip-baited hair traps, and trolled the jungle for the Formosan clouded leopards from 2000 to 2004, spending the time since then to analyze data in an area that was later made impassable by typhoons. The team took 16,000 photos in 400 spots, Pei says. They also looked for paw prints and fur. Still, despite their efforts, they found no trace of the meter-long cats named for their large cloudlike spots.
?There hasn?t been any evidence of their continued existence,? he says.?
The results announced last week confirm a 1985 study that also found no traces of the animal, which is indigenous to Taiwan. Pei says urbanization, farming, poaching, and industry pushed the species from its native lowland hills into poorer habitat in the mountains. The last piece of evidence of their existence here was a picture in a Japanese colonist?s diary dated 1910.
Researchers had held out hope that the species, which is still found in other parts of Asia, could still exist here, too.
The main species of clouded leopards is considered vulnerable to extinction in the?Himalayas. The Bornean clouded leopard, another species, lives on Borneo and Sumatra in Indonesia and is also considered vulnerable.
As much of Taiwan?s public places priority in urban economic successes, few saw animals as relevant to their lives until this news, activists in Taipei say.?
?Awareness of conservation is better than before but still lacking,? says Chu Tseng-hung, executive director of the conservationist group Environment and Animal Society of Taiwan. Reaction often comes down to exposure, he adds. ?With clouded leopards, if you have never seen one, you won?t have any reaction now,? he says.?
Activists hope the clouded leopard case will be an incentive to help save other species here.
Wild animals such as the leopard and Formosan black bear are now threatened by poaching plus development that dominates all but the east coast and high interior mountains. Taiwan, at 634 people per square kilometer, has one of the world?s densest populations. (It?s about the size of the state of Maryland, but has a population of 23 million.) Leopard cats number about 500 and the bears closer to 1,000.?
Whether a groundswell of people will be able to save other species depends on whether activists take this opportunity to get the word out, says Sean McCormack, cofounder of the Taiwan SPCA. ?When the Taiwanese are aware of issues, they get behind them 100 percent,? he says.?
The clouded leopard research team may need support for reintroducing clouded leopards by bringing in a starter population from elsewhere in Asia, Pei says. He hopes to stir up more popular enthusiasm, as the government would need to sign off as well. ?Just for scholars to discuss the issue isn?t that helpful,? Pei says.

TRIPOLI (Reuters) - Libya's parliament voted on Sunday to ban anyone who held a senior position during Muammar Gaddafi's 42-year rule from government, a move which could unseat the prime minister and other top officials regardless of their part in toppling the dictator.
Politicians debated the draft law for months, but the issue came to a head this week when heavily armed groups took control of two ministries and stormed other institutions including the state broadcaster.
The decision to hold the vote under duress could embolden the armed groups to use force again to assert their will over parliament.
Prime Minister Ali Zeidan, a diplomat under Gaddafi who defected to the exiled opposition in 1980, could be among those barred from office, although this remained unclear and a parliament spokesman said it would be decided by a committee charged with implementing the law.
"Being unjust to a few is better than defeating the whole objective of the law," said one of the hundreds who filled Tripoli's main square to celebrating the passage of the law, many of them shooting guns into the air.
Nearly two years after Gaddafi was overthrown, the gunmen who fought to end his 42-year dictatorship are refusing to lay down their arms and go back to civilian life - militiamen are more visible than Libyan state forces in the capital.
The cabinet and Libya's official armed forces are so weak that swathes of the oil-producing desert country remain outside central government control.
A spokesman for parliament admitted it was unclear whether the vote would be enough to dislodge the gunmen from their positions outside the government buildings.
"We hope the siege of the ministries will stop now, but it is not in our hands," General National Congress (GNC) spokesman Omar Hmaiden told a news conference after the vote.
More than a dozen vehicles mounted with anti-aircraft weapons and machine guns remained parked outside the Justice Ministry and the Foreign Ministry has been similarly encircled for a week.
One of the men stationed by a machine gun in front of the Justice Ministry, said the group came from different areas close to the capital Tripoli and ahead of the vote vowed they would stay until the prime minister had been forced from office.
"We have been asking them to deal with Gaddafi's friends for a year," he said.
Although the law passed with an overwhelming majority of 164 votes in favor and just four against, many congress members were upset.
"It's a very unfair and extreme law, but we need to put national interests first in order to solve the crisis," said Tawfiq Breik, spokesman for the liberal National Forces Alliance (NFA) bloc, Libya's largest parliamentary coalition.
Diplomats in Tripoli complained that holding the vote under duress had undermined its legitimacy, while a human rights group called on parliament to reject the draft.
"The GNC should not allow itself to be railroaded into making very bad laws because groups of armed men are demanding it," said Sarah Leah Whitson, a Human Rights Watch director in the region, in a statement.
"Libya's long-term prospects for peace and security will be seriously diminished if the congress agrees to nod through this law."
Much will depend on how high up in Gaddafi's administration an official has to have been in order to be excluded from politics, one analyst said.
"If the bar is too low, the law could result in most government administrations being gutted, without having sufficient staff or institutional memory to ensure their proper functioning," said Geoff Porter of North Africa Risk Consulting.
"However, if the bar is too high then we are likely to see repeats of the blockades in front of government ministries that we saw this week."
Congress members say the law could be applied to around 40 others in the 200-member parliament, including the president of the assembly Mohammed Magarief who became an exiled leader of Libya's oldest opposition movement in the 1980s after serving as an ambassador under Gaddafi.
The law does not make provisions for those, like him, who spent decades in exile and were instrumental in toppling Gaddafi.
The law prohibits former officials from holding any position in government or even belonging to a political party. It will also ban them from leadership roles in the country's state firms, like the National Oil Corporation, its universities and judicial bodies.

May 6, 2013 ? Many high school football players say it's OK to play with a concussion even though they know they are at risk of serious injury, according to a study to be presented Monday, May 6, at the Pediatric Academic Societies (PAS) annual meeting in Washington, DC.
The study of 120 high school football players in the Cincinnati area also found that one-quarter had suffered a concussion, and more than half acknowledged they would continue to play with symptoms of a concussion.
"These attitudes could leave young athletes vulnerable to injury from sports-related concussions," said study co-author Brit Anderson, MD, pediatric emergency medicine fellow at Cincinnati Children's Hospital Medical Center.
Dr. Anderson and her colleagues administered two surveys to the athletes to measure their knowledge of concussions and symptoms as well as their attitudes about playing after a head injury.
Survey results showed that 70 percent of the players had been educated about concussions, and most could identify common signs and symptoms. Headache was identified as a symptom by 93 percent, dizziness by 89 percent, difficulty remembering and sensitivity to light/sound by 78 percent, difficulty concentrating by 76 percent and feeling in a fog by 53 percent.
While 91 percent recognized a risk of serious injury if they returned to play too quickly, only half would always or sometimes report their concussion symptoms to their coach.
"Despite their knowledge, many athletes in our sample reported that they would not tell their coach about symptoms and would continue to play," Dr. Anderson said. "A small percentage even responded that athletes have a responsibility to play in important games with a concussion."
The researchers found no association between a student's knowledge score and attitude score on the surveys. "In other words, athletes who had more knowledge about concussions were not more likely to report symptoms," Dr. Anderson said.
"Although further study needs to be done," she concluded, "it is possible that concussion education alone may not be enough to promote safe concussion behaviors in high school football players."

WASHINGTON (AP) ? As a Senate committee prepares to begin voting this week on far-reaching immigration legislation, advocates are watching warily to see whether opposition, thus far subdued, builds into the same kind of fierce backlash that shut down Congress' last attempt to remake the nation's immigration system.
That time around, in 2007, angry calls overwhelmed the Senate switchboard and lawmakers endured raging town hall meetings and threats from incensed constituents. The legislation ultimately collapsed on the Senate floor.
"I've been through this battle, and it's ugly," said former Sen. Trent Lott, R-Miss., who supported the bill. "My phones were jammed for three weeks and I got three death threats, one of which I turned over to the FBI. So it's rough business."
Supporters of the immigration bill, released last month by a group of four Republican and four Democratic senators, have been cautiously optimistic about their prospects because of factors including public support for giving citizenship to immigrants, a large and diverse coalition in support of the bill, and a growing sentiment among Republican leaders that immigration must be dealt with if they are to regain the backing of Hispanic voters. Backers have been working hard to build alliances and strategies aimed at avoiding the mistakes of 2007, when critics largely defined the bill and some supporters ended up turning against it.
Opponents acknowledge that supporters started out better organized and mobilized than last time around, and they also anticipate that outside groups pushing the legislation ? including efforts headed by New York Mayor Michael Bloomberg and Facebook founder Mark Zuckerberg ? will outspend them. Supporters include large and influential groups including the U.S. Chamber of Commerce, AFL-CIO and the Catholic Church, while opponents include lesser-known think tanks or advocacy organizations such as NumbersUSA, the Federation for American Immigration Reform and the Center for Immigration Studies. Both sides have already begun running ads.
But critics also have important grass-roots influence, including from talk radio hosts who were instrumental in defeating the bill in 2007, and opponents argue that as the public absorbs the content of the legislation, the tide will turn against it. They say that there are already signs that it's happening. Although conservative commentators on Fox News Channel and elsewhere have been more muted so far than in 2007, some talk radio hosts including Mark Levin and Rush Limbaugh have begun to voice deep unease about the bill despite the efforts of its conservative standard bearer, Sen. Marco Rubio, R-Fla., to sell the legislation to them and other conservative opinion leaders.
"The supporters promoted the bill aggressively before anybody saw the language, and certain Republicans and conservative voices sort of held their fire, but that's beginning to change," said Sen. Jeff Sessions, R-Ala., who was a leading voice in the Senate against the bill in 2007 and is reprising that role this time around, making floor speeches, issuing press releases and holding briefing calls with reporters to argue that the bill would unlock a much larger volume of immigration into the U.S. than advertised, to the detriment of U.S. workers and jobs.
"It's going to be like that mackerel in the sunshine ? the longer it's out there the worse it smells," Sessions said.
The bill would aim to boost border security, fix legal immigration and worker programs, require all employers to check their workers' legal status and offer eventual citizenship to the estimated 11 million immigrants already living in the country illegally.
Joyce Kaufman, a host on a Florida radio station, WFTL, said that opposition to the bill was soft at first but grows daily.
"Yes, we believe this is amnesty," Kaufman said. "Citizen activists are outraged."
Lott said that supporters of the legislation still haven't come up with an argument as concise and effective as that one word ? "amnesty" ? from opponents. He said he's spoken with Rubio, among others, to make clear that supporters of the bill need to hone their arguments.
"Last time our explanation was three paragraphs. Theirs was a word," Lott said. When that happens, he said, "You're dead."
The Democratic-led Senate, where the Judiciary Committee takes up the bill on Thursday, is already going to be a tough challenge. But if the bill does pass the Senate, opponents are betting it gets stopped in the Republican-led House. A bipartisan group of House lawmakers has been promising for months to release their own bill mirroring elements of the Senate legislation but taking a tougher tack. So far they haven't delivered.
Meanwhile, to the dismay of immigration advocates, the chairman of the House Judiciary Committee has announced plans to move forward with individual, single-issue immigration bills, rejecting the comprehensive approach in the Senate that's backed by President Barack Obama, who's made immigration legislation a top second-term priority. The legislation was also a priority in 2007 for then-President George W. Bush, but he was unsuccessful in persuading Republican lawmakers to get behind the bill, and Democrats who at the time controlled Congress were divided, too.
In the 2007 debate, a turning point came when the conservative Heritage Foundation released a report saying that the legislation would cost taxpayers $2.6 trillion, including benefits to immigrants and other expenditures. Although the analysis was disputed it carried weight with GOP lawmakers. Now under the leadership of former Sen. Jim DeMint, R-S.C., another lead opponent of the legislation in 2007, Heritage is preparing to release an updated version of that report.
In a sign of how supporters of the bill are working hard not to repeat mistakes from the past, conservative groups that support the legislation have already sought to pre-empt the Heritage report, with the Cato Institute deriding it ahead of time as "fatally flawed," and Cato and others arguing that immigration reform would boost the economy by growing the labor market. Nonetheless officials with Heritage argue their report could have the same impact this time around as in 2007.
"There's been a lot of posturing, a lot of talk. We haven't really gotten to the heart of the debate yet," said Dan Holler, communications director for Heritage Action for America, the Heritage Foundation's activist arm. "We have the right policy, the numbers are going to be there, and the debate is going to shift. And no amount of ads will be able to shift it back."

The U.S. Department of Energy Joint Genome Institute (DOE JGI) is among the world leaders in sequencing the genomes of microbes, focusing on their potential applications in the fields of bioenergy and environment. As a national user facility, the DOE JGI is also focused on developing tools that more cost-effectively enable the assembly and analysis of the sequence that it, as well as other genome centers, generates.
Despite tremendous advances in cost reduction and throughput of DNA sequencing, significant challenges remain in the process of efficiently reconstructing genomes. Existing technologies are good at cranking out short fragments (reads) of DNA letters that are computationally stitched back together (assembled) into longer pieces, so that the order of those letters can be determined and the function of the target sequence discerned. However, genome assembly, the equivalent of trying to put together a multi-million piece jigsaw puzzle without knowing what the picture on the cover of the box is, remains challenging due to the very large number of very small pieces, which must be assembled using current approaches.
As reported May 5 online in the journal Nature Methods, a collaboration between the DOE JGI, Pacific Biosciences (PacBio) and the University of Washington has resulted in an improved workflow for genome assembly that the team describes as "a fully automated process from DNA sample preparation to the determination of the finished genome."
The technique, known as HGAP (Hierarchical Genome Assembly Process), uses PacBio's single molecule, real-time DNA sequencing platform, which generates reads that can be up to tens of thousands of nucleotides long, even longer than those provided by the workhorse technology of the Human Genome Project era, the Sanger sequencing technology, which produced reads of about 700 nucleotides. The Sanger process involved creating multiple DNA libraries, conducting multiple runs, and combining the data, so that gaps in the code were covered and accuracies of a DNA base assignment were very high. Post-Sanger methods still typically require multiple libraries and often a mix of technologies to produce optimal results. Instead, with HGAP, "only a single, long-insert shotgun DNA library is prepared and subjected to automated continuous long-read SMRT sequencing, and the assembly is performed without the need for circular consensus sequencing," the team reported.
This de novo assembly method was tested using three microbes previously sequenced by the DOE JGI. The data collected were compared against the reference sequences for these microbes and the team found that the HGAP method produced final assemblies with >99.999% accuracy.
"We are always on the lookout for new approaches that will improve upon the efficient delivery of high-quality data to our growing community of researchers," said Len Pennacchio, DOE JGI's Deputy Director of Genomic Technologies. "This technique is one of many improvements that we are pursuing in parallel to achieve additional economies of scale."
The DOE JGI's sequencing efforts account for more than 20% of the more than 20,000 worldwide genome projects (microbes, plants, fungi, algae, and communities of microbes) completed or currently in the queue, and most of those are focused on the biology of environmental, energy, and carbon processing.
"We enjoyed a very productive collaboration with JGI on this project and benefited tremendously from the expertise of JGI's scientists in both the fields of microbiology and microbial genome assembly and annotation," said Jonas Korlach, Chief Scientific Officer at Pacific Biosciences. "This expertise provided us with the ability to adapt our single molecule sequencing assembly methods to produce a higher level of finished quality than was previously possible using a gold-standard Sanger finishing approach, and at a speed and price point competitive with alternative next generation sequencing and assembly methods. We look forward to seeing what scientific advances will be enabled by this method as JGI's User Community assesses JGI's capabilities to assemble their microbial genomes using this new approach."
The team will now seek to extend the utility of this new assembly method beyond microbes to the genomes of more complex organisms.

May 4, 2013 ? As parents, physicians and policymakers look for ways to curb childhood obesity, they may need to look no further than a child's own backyard.
A new study to be presented Saturday, May 4, at the Pediatric Academic Societies (PAS) annual meeting shows that preschool children are less likely to be obese if they live in a neighborhood that is safe and within walking distance of parks and retail services.
"A child's neighborhood is a potentially modifiable risk factor for obesity that we can target in order to stop the increasing prevalence of obesity in young children," said lead author Julia B. Morinis, MD, MSc, a pediatrician at Hospital for Sick Children in Toronto, Ontario, Canada.
The study is part of a Canadian research collaboration called TARGetKids! (The Applied Research Group for Kids) that aims to determine if factors early in life are related to later health problems. Healthy children ages 0-5 years are enrolled in the ongoing study. Information is collected on their height, weight, waist circumference, nutrition, and physical and sedentary activity. Blood samples also are taken from each child.
For this study, researchers used TARGetKids data on 3,928 children in Toronto to determine if where they live was related to whether they were overweight or obese. Neighborhoods were evaluated based on car ownership, population, distance to retail locations, distance to parks and safety.
Results showed that 21 percent of the children were overweight, and 5 percent were obese, which is similar to the Canadian norms. Higher rates of overweight/obesity were found among children who live in neighborhoods that have fewer destinations within walking distance.
"How conducive a child's neighborhood is to physical activity is related to a child's body mass index (BMI) even after adjusting for factors we know are associated with obesity, including socioeconomic status, immigration, ethnicity, parental BMI, physical activity, age, gender and birth weight," Dr. Morinis said.
Further research is needed to better understand the relationship between neighborhood factors and obesity so that the risk of obesity can be reduced through neighborhood changes and urban planning, the investigators concluded.

Behind the net gain of 165,000 jobs last month was an improved housing industry, a decline in the price of gasoline that has helped consumers, and a resurgent stock market that may have made some people feel richer.????
?The jobs numbers certainly suggest that things are not falling off the cliff,? says Scott Brown, chief economist at Raymond James & Associates in St. Petersburg, Fla. ?It looks like the spring slump was less severe than prior years.?
Not only was April report better than expected, but the US Bureau of Labor Statistics also revised upward February's and March's numbers by a total of 114,000 jobs. The upward revisions meant that in March employment grew by 138,000 instead of the first-reported 88,000, and in February jobs increased to 332,000 from 268,000.
The stock market seemed to be encouraged by the jobs numbers, perhaps because they seem to indicate that the Federal Reserve is not likely to make any policy changes in the months ahead. As of noon, the Dow Jones Industrial Average is up 156 points to 14987, after briefly going over 15000 for the first time ever. ?
However, economists caution that the jobs numbers do not signify a vibrant economy. The data also showed a drop in the average number of hours people worked per week ? perhaps indicating that orders are slowing, causing businesses to cut back on overtime and add more part-time workers. In addition, economists anticipate that the effect of recent federal budget cuts will increase as summer approaches.
?In terms of the general economy, the growth of jobs is just OK,? says Dan Meckstroth, chief economist at the Manufacturers Alliance for Productivity and Innovation in Arlington, Va. ?It is just above the 120,000 new jobs needed each month to absorb the new entrants into the economy.?
As economists parsed the April data, some were particularly troubled by a fall in the work week by 0.2 hours. In an analysis sent to clients, Marc Chandler, global head of currency strategy at Brown Brothers Harriman, estimates that this reduced the average work week by 12 minutes for the 135 million labor force.
Mr. Chandler says the reduction in the average work week appears to be a function of an increase in the number of part-time jobs. ?The number of people ?involuntarily? taking part-time work rose 278,000 to 7.64 million,? he wrote.
However, at the same time, the BLS reported the number of people taking ?temp? jobs rose by 31,000 last month. According to temp-service executives, a rise in temp jobs may presage a later rise in full-time jobs, as employers become more confident about adding workers.
The BLS also reported an increase in other white-collar jobs in April, notably a gain of 73,000 jobs in professional and business services. Since the beginning of the year, this category has added 587,000 jobs.
Other service-sector jobs also appeared to be growing. The BLS reported a gain of 38,000 jobs in leisure and hospitality. Over the prior 12 months, this category has added an average of 25,000 new jobs per month.
Manufacturing jobs, however, were flat in April, according to the BLS. Mr. Meckstroth says business is not being helped by a weakening economy in Europe and slower growth in Asia.
However, the improvement in new home construction has continued to add jobs. Over the past six months, construction has averaged 27,000 new jobs per month, says the BLS.
?There are still shortages in skilled workers, machinists, welders, the skilled occupations,? says Meckstroth. ?Unfortunately, labor force skills don?t match up with demand.?
The demand for skilled workers is likely to continue for some time.
There is a significant pent-up demand for new automobiles, for example. This week, Ford Motor Co. said it would hire 2,000 workers at its Claycomo, Mo., plant, which makes F-150 pickup trucks. Of the new hires, 900 will work on a third shift.
Demand also is growing for people who can help build new homes.
Meckstroth estimates that new housing starts will jump 24 percent this year and 34 percent next year. Although they are rising from very low levels, he notes that it is having a spill-over effect on demand for wood products, appliances, structural metals, gypsum, and heating and air-conditioning systems.
?It is feeding through all the other materials that go into making a house,? he says. "Unfortunately, it?s not enough to offset weaknesses in other areas.?
Government jobs continued to shrink last month with a loss of 11,000 workers. Of these, 3,500 were part of the shrinking US Postal Service. Loss of jobs at the postal service over the past year has helped to bring total federal government employment down to its lowest level since December 2008.

LONDON (Reuters) - Tax havens such as Bermuda and the Cayman Islands will work more closely with Britain and other European countries to fight tax evasion, Chancellor George Osborne said on Thursday.
With governments in most advanced economies short of tax revenue after the financial crisis, pressure has been growing on small territories with big banking sectors to lift bank secrecy and do more to combat tax dodging and money laundering.
The United States has been in a lengthy dispute with Switzerland over the latter's bank secrecy rules, while the European Union also has turned up pressure on two of its smaller members, Austria and Luxembourg.
Austria in turn has accused Britain and the United States of shielding their own tax havens.
Now British overseas territories such as Bermuda, the Cayman Islands and the British Virgin Islands - which are effectively colonies with some self-government - will have to automatically provide details the ownership of bank accounts, and about how they are used, the Treasury said.
"This represents a significant step forward in tackling illicit finance and sets the global standard in the fight against tax evasion," Osborne said.
The information will be shared by Britain, Germany, France, Italy and Spain, which agreed in June last year to work together to combat tax evasion.
The Isle of Man also will be subject to the deal, but two other major offshore centres, Jersey and Guernsey, have not yet agreed to share information with countries other than Britain.
The other territories affected by the deal are the Caribbean islands of Anguilla, Montserrat and the Turks and Caicos.
Britain has made fighting tax evasion a priority while it chairs the G8 group of advanced economies this year, and in two weeks it will host a meeting of finance ministers from the bloc.
However, Britain also faces problems of its own in being certain that companies comply with its tax laws.
A Reuters investigation published on Wednesday highlighted inconsistencies in the way Google portrays its activities in Britain, prompting parliamentarians to demand the company's executives and auditors appear before them.

OAKLAND, Calif. (AP) ? The NBA fined Golden State coach Mark Jackson $25,000 on Thursday for making public comments in an attempt to influence the officiating in the Warriors' playoff series with Denver.
After Golden State's 107-100 loss in Game 5 at Denver on Tuesday night, Jackson said the Nuggets sent "hit men" and took "cheap shots" at point guard Stephen Curry. He repeatedly mentioned Kenneth Faried's foul in the first half, when the forward stuck out his right leg and moved his body while Curry ran through traffic.
"For two years now, I've taken pride in not ever criticizing referees ? for two years. And then reading the statement by the NBA, I'm extremely thankful I am not fined for criticizing referees," Jackson said before Game 6 against the Nuggets on Thursday night.
Told the league fined him for an "attempt to influence the officiating," Jackson responded: "I don't like it, and I disagree." He declined to say whether he would appeal the fine.
Jackson, who doubles as an ordained minister, had never been fined for criticizing officials in his two years as an NBA coach. He couldn't recall being fined for criticizing officials in his 17 season as a point guard in the league, but said his team wouldn't allow all the rhetoric ? or his fine ? to be a distraction.
"This is an awesome time. I truly don't take it for granted," Jackson said. "I smile, $25,000 lighter, but I'm smiling."
Jackson also questioned how Denver might try to defend Curry before Wednesday's practice.
"They'll be physical again. They'll try to beat up Steph Curry. They'll try to set illegal screens. They'll try to chuck him when he goes down the lane," said Jackson, whose team took a 3-2 series lead into Game 6. "Other than that, I'm not sure what to expect out of them."
Denver Nuggets coach George Karl thought the reaction to his team's Game 5 victory sounded like a "different movie than I'm watching." Karl also said: "I'll go to any arbiter right now and show the dirty shots. They're winning."
